Rapid Coomassie Blue Protocol

Remove Gel

Turn off power to gel.

Take out core and drain core into sink.

Remove gel and glass plate sandwich from central core.

Take off clamps.

Slide one spacer out gently from between the 2 glass plates.

Twist spacer to separate plates from the gel.

Take off the other spacer.

Remove the plate the gel isn't most attached to.

Cut away the stacker portion of the gel and discard.

Use a razor blade to pull up one corner of the gel and pull gel off of the plate as gently as possible.

 

Fixing the gel:

Put the gel in Isopropanol Fixing Solution on orbital shaker for 30-60 minutes.

Isopropanol Fixing Solution: Stock For 1 Liter

a. 25% Isopropanol 100% 250mls

b. 10% Acetic Acid 100% 100mls

c. 65% H2O 650mls

Dump out fixing solution by holding one corner of the gel and dumping the fix into the sink

Put the gel in Isopropanol Fixing Solution on orbital shaker for 30-60 minutes.

Isopropanol Fixing Solution: Stock For 1 Liter

a. 25% Isopropanol 100% 250mls

b. 10% Acetic Acid 100% 100mls

c. 65% H2O 650mls

Dump out fixing solution by holding one corner of the gel and dumping the fix into the sink.

Staining

Put the gel in Isopropanol Fixing Solution on orbital shaker for 30-60 minutes.

Isopropanol Fixing Solution: Stock For 1 Liter

a. 25% Isopropanol 100% 250mls

b. 10% Acetic Acid 100% 100mls

c. 65% H2O 650mls

Dump out fixing solution by holding one corner of the gel and dumping the fix into the sink

Cover the gel with Rapid Coomassie Blue Staining Solution.

Coomassie Blue Staining Solution: Stock For 1 Liter

a. 10% Acetic Acid 100% 100mls

b. 0.006% Coomassie Brilliant Blue 0.06g

c. 90% H2O 900mls

Put on shaker and stain until gel reaches desired intensity.

Stain 2 hours to overnight covered at room temperature.

Dump out staining solution.

De-Stain:

Put the gel in Isopropanol Fixing Solution on orbital shaker for 30-60 minutes.

Isopropanol Fixing Solution: Stock For 1 Liter

a. 25% Isopropanol 100% 250mls

b. 10% Acetic Acid 100% 100mls

c. 65% H2O 650mls

Dump out fixing solution by holding one corner of the gel and dumping the fix into the sink

Cover the gel with 10% Acetic Acid to de-stain.

Put on orbital shaker 2 hours or more at room temperature until a clear background is obtained.

Dry Gel:

Put the gel in Isopropanol Fixing Solution on orbital shaker for 30-60 minutes.

Isopropanol Fixing Solution: Stock For 1 Liter

a. 25% Isopropanol 100% 250mls

b. 10% Acetic Acid 100% 100mls

c. 65% H2O 650mls

Dump out fixing solution by holding one corner of the gel and dumping the fix into the sink

Put gel in 40% methanol, 10% Glycerol, and 7.5% Acetic Acid solution for 3-5 minutes.

Gel Drying Solution: Stock For 1 Liter

a. 40% Methanol 100% 400ml

b. 10% Glycerol 100% 100ml

c. 7.5% Acetic Acid 100% 75ml

d. H2O 425ml

Moisten 1 sheet of gel drying film in gel drying solution. Do not keep film in solution for more than 1 minute.

Lay one gel drying frame down flat on the bench and put a glass plate in the middle to help make the film even.

Lay film over gel drying frame and remove any wrinkles.

Roll gel onto film and remove any bubbles (bubbles may cause cracking in gel).

Moisten another piece of gel drying film in gel drying solution.

Roll film onto frame and gel, one end at a time to avoid bubbles.

Lay second frame down onto other frame.

Clamp the frames together by pulling one side of the frame off the bench, then fold over the gel drying film and clamp together. Clamp with 3 clamps per side.

Stand clamped frames up in hood and let dry 24-48 hours.

Remove clamps and cut away the extra film.

Tape into notebook and label gel.
