Initializing Nikon Microscope

Hook up light pen to back of scope, turn on power.

Scan: Initialize

Scan: Version 1.1-à3 beeps= newer version than 1.1 installed

Set Home position.

            Move stage down till beep is heard

            Scan: Home position

Set focus position

            Make sure Eyepieces are "zeroed"

Using 10X obj. move stage up till sample is in focus

            Scan: Reference Focal Position Setting

Identify objectives

            Scan: Data setting address 1

            Scan: plan apo 4x (or whatever objective is in position 1.

            Repeat this for each position.

            Nothing in a position, don't need to do anything

Identify objective turret (Nosepiece)

            Scan: motorized           

            Scan: 7 place

Set Focus offset/parafocalizing

            Using 4X obj. focus on sample

            Scan: Focal position setting

            Repeat with all objectives

Reboot, ie turn power on then off

Set objective selector switch if desired- this allows you to run filter cubes from side switches. Default is that both sides run objective turret.

 Kohler Illumination/ Field Diaphragm Adjustment

            Adjust condenser up/down till octagonal edges of field diaphragm are in sharp focus.

            Center the field diaphragm using adjuster knobs located on condenser, one on each side.

            Open field diaphragm (on bottom right side) till diaphragm is just outside field of view
